Research Interests

The primary goal of my research is to determine the molecular basis of the biological function of the protein, human relaxin, and to use the resulting information to prepare domain-minimised mimetics having either agonist or antagonist actions for use as potential therapeutics. There are two human relaxins known as relaxins-2 and -3. The former has clear roles in parturition and tissue remodelling. Relaxin-2 acts to regulate collagen synthesis and is proving to be very useful in reducing kidney and lung fibrosis. Additionally, over-expression of relaxin-2 has been associated with breast, prostate and thyroid cancers. In contrast, relaxin-3 is produced in the brain and is rapidly emerging as a key neuropeptide that seems to be involved in appetite and stress regulation. It has been shown to stimulate feeding and thus may have important therapeutic applications in cases of anorexia. Conversely an antagonist of the peptide may be a potential anti-obesity agent.

To date, I have exploited my considerable expertise in the solid phase chemical synthesis of these complex two chain, three disulfide bonded peptides to prepare numerous analogues of relaxins-2 and -3 for structure-function relationship mapping via NMR spectroscopic tertiary structural determination and receptor assay. My work has led to a clear elucidation of the active sites of the two peptides. The sites are structurally similar despite the two peptides acting on different receptors. I am now using the resulting information together with computer-aided drug design techniques to prepare domain-minimised analogues of the relaxins with therapeutically useful agonist or antagonist activities.
